Amgen’s Translational Safety & Bioanalytical Sciences department at our Thousand Oaks location is involved in the development and establishment of methods capable of detecting binding and neutralizing anti-drug antibodies (ADAs) to therapeutic proteins in clinical samples. Interference from circulating drug present in the test samples is a critical challenge for these methods for the reliable detection of ADAs. Currently the most common assay format for detection of anti-drug antibodies (ADA) in clinical studies is the Bridging Assay. In this assay, the bivalent nature of ADA enables the formation of a bridge between labeled drug molecules, resulting in assay signal. Although this format is highly sensitive, it has several important limitations: 1) false positives due to the presence of non-ADA molecules, such as multimeric drug target, 2) false negatives due to the presence of high levels of circulating drug, and 3) requirement for drug labeling. Alternative formats have the potential to overcome these challenges but cannot be used for therapeutics that contain human Fc because this region is shared between the capture reagent (therapeutic) and the analyte (ADA). We are currently evaluating new approaches to solve this problem.

 

The intern will work with the project scientist to explore these approaches and/or develop a novel approach for ADA assays and aim to complete the evaluation in 2-3 months.

 

Basic Qualifications

 

Amgen requires that all individuals applying for a grad internship or a co-op assignment at Amgen must meet the following criteria: 

 

18 years or older

 

Graduated with a Bachelor’s degree from an accredited college or university with a 3.0 minimum GPA or equivalent

 

Completion as a first year graduate student before internship starts and currently enrolled in a Master’s program for a Masters internship OR a PhD for a PhD internship from an accredited college or university

 

Enrolled in an accredited college or university following the potential internship or co-op assignment

 

Must not be employed at the time the internship starts

 

Preferred Qualifications

Pursuing PhD in Life Sciences

 

In at least the third year of current program

 

Experience with ELISAs and data analysis using Excel

 

Working knowledge of immunology
